The insert contains the following sites in kb from the vector SmaI site (or the published sequence): SstI--0.2 (+666), HindIII--0.6 (+289), EcoRI--0.7 (+197), BglII--2.2 (-1443), BglII--2.8, SphI--3.8, EcoRI--5.7, SphI--6.0, SstI (7.1). Restriction digests of the clone give the following sizes (kb): SmaI/BglII--13.0, 2.8, 0.8; SstI--9.4, 8.7; BglII--16.0, 0.8; EcoRI--6.5 (doublet), 2.3 (doublet). The ATP10 gene product is 30.3 kDa and appears to be required for the assembly of the F1-F0 ATPase. atp10 mutations decrease the rutamycin sensitivity of the ATPase. The ATP10 sequences are contained in a 2.2 kb SmaI/BglII fragment which contains some sequences from the vector. This fragment also contains 2 other open reading frames. |
